Involvement of the Kv10.1 potassium channel expressed by the breast cancer cells in the development of brain metastasis

Breast cancer is a carcinoma, this type of cancer is developing breast tissue in women. There are different types of breast cancers ; luminal A, luminal B, triple negative then HER2-enriched. Those types induce different metastasis sites in patients and are not equivalent about prognosis. In fact, triple negative breast cancer is the worst case possible with a great aggressiveness and multiple secondary sites like lymphatic ganglia, lungs or brain. My internship focuses on brain metastases and its invasion mechanisms with a cell line of breast cancer known to be invasive ; MB-MDA-231. A promising target in carcinogenesis is Kv10.1, a potassium channel. Indeed, this channel was shown to promote migration and invasion of breast cancer, but its potential function in brain invasion is not known. In the Laboratoire de la Barrière Hémato-Encéphalique (LBHE) in Lens, I am going to study with blood-brain-barrier cellular model the Kv10.1 in migration, invasion and endothelial permeability then in the Laboratoire de Physiologie Cellulaire et Moléculaire (LPCM) in Amiens is planned to check up some actors (secretome) involved in response to silencing Kv10.1.
